Comparative genomics of closely related Thermococcus isolates, a genus of hyperthermophilic Archaea ArchiMer

The immense genomic diversity of microorganisms allows them to live everywhere, even in extreme environments such as deep hydrothermal vents. Scattered over the seabed, these are a good model for studying the biogeography and genomes diversification. A comparative genomics approach has been used on closely related isolates, of the genus Thermococcus. This work aimed at identifying mechanisms that have a role in the diversification of these genomes, and also to identify genes involved in this differentiation. Draft genome sequence of sulfur-reducing archaeon Thermococcus thioreducens DSM 14981T BJM

Abstract Thermococcus thioreducens DSM 14981T, a sulfur-reducing archaeon, was isolated from the rainbow hydrothermal vent site on the Mid-Atlantic Ridge. Herein, we report the draft genome sequence of T. thioreducens DSM 14981T; we obtained 41 contigs with a genome size of 2,052,483 bp and G + C content of 53.5%. This genome sequence will not only help understand how the archaeon adapts to the deep-sea hydrothermal environment but also aid the development of enzymes that are highly stable under extreme conditions for industrial applications.

Since the discovery of hydrothermal vents in 1977, a small number of studies have allowed the isolation and characterization of microorganisms that can withstand high pressures and temperatures. Among these, Pyrococcus yayanosii, a hyperthermophilic archaea of the order Thermococcales, came from the Ashadze site (Mid-Atlantic Ridge) at 4100 m depth. This species represents the first organism both hyperthermophilic and strict piezophilic described to date. It cannot divide at pressures below 20 MPa and its optimal growth pressure is 52 MPa. Physiology and continuous culture of the hyperthermophilic deep-sea vent archaeon Pyrococcus abyssi ST549 ArchiMer

The deep-sea vent archaeon Pyrococcus abyssi strain ST549 was grown in batch cultures in closed bottles and by continuous culture in a gas-lift bioreactor, both in the presence and in the absence of elemental sulfur. Growth on carbohydrates, proteinaceous substrates and amino acids was investigated. The disaccharides maltose and cellobiose were shown not to be able to enhance growth suggesting that P. abyssi ST549 is unable to use them as carbon sources. By contrast, proteinaceous substrates such as peptone and brain heart infusion were shown to be very good substrates for the growth of P. abyssi ST549 and allowed growth at high steady-state cell densities in continuous culture. The molecular organization of the replication complex in archaea is similar to that in eukaryotes. Only two proteins homologous to subunits of eukaryotic replication factor C (RFC) have been detected in Pyrococcus abyssi (Pab). The genes encoding these two proteins are arranged in tandem. We cloned these two genes and co-expressed the corresponding recombinant proteins in Escherichia coli. Two inteins present in the gene encoding the small subunit (Pab RFC-small) were removed during cloning. The recombinant protein complex was purified by anion-exchange and hydroxyapatite chromatography. The PCNA from Thermococcus fumicolans functionally interacts with DNA polymerase delta ArchiMer

We have cloned the gene encoding proliferating cell nuclear antigen (PCNA) from the hyperthermophilic euryarchaeote Thermococcus fumicolans (Tfu). Tfu PCNA contains 250 amino acids with a calculated M-r of 28,000 and is 26% identical to human PCNA. Next, Tfu PCNA was overexpressed in Escherichia coli and it showed an apparent molecular mass of 33.5 kDa. The purified Tfu PCNA was tested first with recombinant Tfu DNA polymerase I (Tfu pol) and second with calf thymus DNA polymerase delta (pol delta). When tested with the homologous Tfu pol on bacteriophage lambda DNA, large amounts of Tfu PCNA were required to obtain two- to threefold stimulation.
